Use of sperm densitometer to measure sperm concentration：
：Choose two colorimetric bottles. We added 10 ml dilution solution to one of the colorimetric bottles as reference color bottle. For another colorimetric bottle, 0.4ml is added through the pipette to activate the original semen and 10 ml diluent solvent is used as the colorimetric bottle. And mix it evenly.
：Opening the sperm densitometer through the button (ON/OFF). Use the reference colorimetric bottle (that is, without adding the original semen solvent) to clear the sperm densitometer and press the (ZERO) key. You can see it on the screen (0).
：Remove the reference color bottle. Measurement of a colorimetric bottle with a solvent of semen. A numeric value can be obtained by pressing the button (READ/DIRECT).
：According to the conversion table. The value obtained from the instrument corresponds to the table. Get the value of the concentration. Relative to the volume of the liquid we use. We can get the amount of sperm in the semen.
：There may be a number of abnormal shapes of spermatozoa in the sperm (for example, rolling up the tail, decreasing the cytoplasm, and being damaged). We can see through a microscope. Operation: first, we use a pipette to extract a drop from the solvent with sperm. It was placed in the slides of the microscope for observation. The number of abnormal sperm counts. The abnormal shape of sperm can be obtained by dividing the number of sperm that is abnormal shape by taking out the volume of the liquid. The concentration of sperm in the abnormal shape is reduced by the concentration in the conversion table. The normal shape of sperm concentration can be obtained. The normal shape of sperm can be obtained by multiplying the concentration and volume of the normal shape sperm.
